iPSC and Organoids

GENERAL REGULATIONS


  • The iPSC and Organoids Laboratory is exclusively dedicated to the generation and culture of iPSC lines and their derived organoids.


  • To request a specific technical service, the user must send, via email to aartero-ibis@us.es, the corresponding form available on the IBiS server notice board (Solicitud_iPSCyORGANOIDES_Anexos).


  • Due to the complexity of the experiments, users must contact the Service to design the experiment and request a quote via email (aartero-ibis@us.es).


  • Only users authorised by the Principal Investigator and who have previously submitted a request to the Service Technician may access and use the iPSC and Organoids Laboratory. The request must be made by sending the corresponding form, available on the IBiS server notice board (Annex 1), to aartero-ibis@us.es.


  • Within the iPSC and Organoids Laboratory, there will be only one IVF cabinet available for those users authorised by the Platform Technician. Priority will be given to users generating iPSCs as a service of the platform, for the maintenance of iPSCs, once they have completed the required hands-on practical training for this type of culture.


  • Users MUST ALWAYS reserve the cabinet they intend to use, for the entire estimated working time. Reservations must be made through the IBiS Booking System, selecting the iPSC and Organoids room and the corresponding cabinet.


  • No user may access the Service without first being familiar with its Regulations, which are available on the IBiS server notice board (Normativas).


  • Access to the iPSC and Organoids Laboratory will be controlled by an electronic system. Each user will be issued a personal card allowing entry and exit.


  • The card is PERSONAL and NON-TRANSFERABLE. Misuse will result in deactivation, and a new access card may not be requested until one month after the incident.


  • Users MUST ALWAYS swipe their card upon entering and leaving the laboratory, even if the door is already open.


  • To enter the iPSC and Organoids Laboratory, it is MANDATORY to use the following items, available at the laboratory entrance:


- Disposable lab coat.

- Protective gloves certified for biological risk (EN ISO 374-5).

- Disposable shoe covers.


  • To work in the IVF cabinets, it is MANDATORY to use:


- Disposable lab coat.

- Surgical mask.

- Extra-long protective gloves certified for biological risk (EN ISO 374-5).

- Disposable cap.

- Disposable shoe covers.


  • The door of the iPSC and Organoids Laboratory must remain closed at all times to ensure optimal sterility and containment.


  • Users must keep the iPSC and Organoids Laboratory in perfect condition regarding cleanliness and sterility. The Service Supervisor will inspect the facilities daily, notifying any incidents to users’ Principal Investigators (PIs). If the issue persists, it will be reported to Management.


  • Users may use ONLY the cabinet and incubator assigned for external users. All other cabinets and incubators are for the EXCLUSIVE use of Service Technicians.


  • Users must collect and store all their materials in the designated area assigned by the Service Supervisor, who will dispose of any improperly stored items.


  • The transport of biological material between the iPSC and Organoids Laboratory and other laboratories or facilities within the IBiS building must be carried out in hermetically sealed containers, which must be disinfected before and after use.


  • Users must periodically clean their assigned areas to avoid the accumulation of unnecessary items.


  • Users must ensure that waste containers are not overfilled. In the case of the yellow biohazard container, it should be sealed and placed inside the green biohazard container, replacements for which are stored in one of the laboratory’s culture compartments. The user who fills the green container last is responsible for sealing it and placing it in the corridor next to the corresponding laboratory door before 11:00 a.m. for collection by Cleaning Services. If this deadline is missed, the sealed container must remain inside the culture lab and be replaced the following day.


  • Users must also periodically clean the contents of refrigerators and freezers to remove expired or unused reagents. The Service Supervisor will assign storage space at 4°C and -20°C, and will dispose of any improperly stored materials. When necessary, the Service Technician will coordinate freezer defrosting with users to ensure the orderly transfer of materials.


  • Users must ensure that all equipment—including cabinets, vacuum pumps, microscopes, centrifuges, and water baths—is turned off if no other user remains in the room to use them.


  • It is strictly forbidden to leave the culture area wearing PPE specifically designated for this room (lab coat, gloves, mask, cap, shoe covers).


  • The use of mobile phones within the culture area is strictly prohibited.


  • The introduction of food or drinks into the culture area is strictly prohibited.
